Dhawan shared 253 runs for the second wicket with Cheteshwar Pujara, who was batting on 75 after the tourists won the toss and elected to bat. This was India's second highest partnership against Sri Lanka in Tests.
The southpaw added 126 runs in the second session to become second in the list of Indians who have scored maximum runs in one session. Virender Sehwag tops the list with 133 runs in 2009 against Sri Lanka.
Dhawan was dismissed just before tea when he offered a simple catch to Angelo Mathews off paceman Nuwan Pradeep.
Earlier, Asela Gunaratne was ruled out of the first Test with a fractured left thumb.
Top-ranked India handed a Test cap to all-rounder Hardik Pandya and Sri Lanka brought in debutant Danushka Gunathilaka for the game.
